Annotator commentaries

Nous avons un ORF qui est codant. Nous avons choisit par défaut l'ORF le plus long. Il est donc dans le sens direct. Le poids moléculaire potentiel serait de 25.56kD

L'analyse des domaines protéique indique que notre ORF coderait pour une peptidase M41. Celle ci intervient dans le catabolisme des protéines. Elle a une activité peptidase qui de plus est ATP dépendante.

Lors de la comparaison de notre ORF avec la banque de donnée " nr " on constate que la quasi totalité des séquences présentant un score de plus de 200 sont des Protéobactéries (en particulier des alpha et gammaProtéobactéries ).On peut donc émettre l'hypothèse que l'ORF étudiée proviendrait d'une protéobactérie et plus probablement à une alphaprotéobactérie . De plus nous pouvons voir que notre ORF est relativement proche de deux candidatus pelagibacter ubique(htcc1002/1062) qui sont des rickettsies. De plus les deux autre homologues suivants sont aussi des rickettsies.Nous pouvons emettre l'hypothèse que notre orf serait une rickettsie. Notre ORF serait probablement un proche cousin de candidatus pelagibacter. Notre groupe d'étude est l'alphaprotéobactérie(de façon plus générale les protéobactéries )et notre groupe extérieur est composé de GNSbactérie ,de GSbactérie et de Eubactérie . Tous les orthologues à l'ORF étudiée codent soit pour des métalloprotéases ATP dépendante ou des peptidase M41 ou bien pour des protéines qui interviennent dans la division cellulaire .Dans tous les cas de figure toute ces protéines interviennent tous durant la division cellulaire . L'ensemble de ces orthologues ont le même symbole Gène il s'agit de " FtsH ", nous proposons donc ce même Symbole Gène pour notre ORF. De plus, après vérification dans la base de donnée swissprot, nous pouvons dire que le gène FtsH intervient lors de la division cellulaire: c'est une protéase de la division cellulaire. Donc ou peut penser que notre ORF coderait lui aussi pour une protéase de la division cellulaire.

Si on fait un BLASTp mais cette fois ci avec la banque de donnée SWISSPROT, on observe que les séquences protéiques qui ont un score les plus élevés sont des protéases intervenant dans la division cellulaire .On a préféré travaillé sur "nr" car le nombre de séquences est beaucoup plus importante et les scores sont meilleures ainsi que les E-Values.

On peut rajouter d'après l'arbre phylogénétique que notre ORF est très proche de deux alpha protéobactéries: candidatus pelagibacter ubique HTCC1002 et HTCC1062.

Nous voulons rajouter qu'on a raciné l'arbre avec GNS bactérie .

Pour aller plus loin voici ce que nous savons à l'heure actuelle sur les candidatus pelagibacter: Pelagibacter ubique est une petite alphaproteobacteria hétérotrophe, trouvée dans tous les océans. C'est peut être la bactérie la plus représentée au monde (peut être 10puissance28 cellules). Cette bactérie était à l'origine nommée SAR11 et connue seulement grâce à ses ARNr identifiés d'échantillons de la mer des Sargasses en 1990. La bactérie a ensuite été isolée et décrite en 2002.

Pelagibacter a une distribution mondiale, elle est un composant du bactérioplancton. Très petite, elle a une longueur de 0,37-0,89 µm et un diamètre de seulement 0,12-0,20 µm.

Tout petit, le génome de Pelagibacter est séquencé : il contient 1,3 Mpb et 1 354 gènes. Il n’y a pas d’introns, pas de transposons, pas de pseudogènes, pas d’éléments extragéniques, pas d’intéines et peu de paralogues.
